OurBus is now offering Ottawa-Toronto trips. Street stops at both termini, with pickup/dropoff also available in Whitby and Kingston.
The ticket price is shown as $9.52. However, a "booking fee", "fuel surcharge fee", and "facility fee" are all added, making the actual price $19.95. Airlines are no longer allowed to do this sort of pricing structure in Canada, but AFAIK there's nothing preventing a bus service from doing so. Kinda scummy though ..read more

Just had this company show up on my radar. Corporate name is Intercity Bus, Inc and they have a valid CVOR: 214669917
Seems they bought over the old Greyhound Canada Bus Terminal in London and their first Route will be London (Downtown) - London (Western University) - London (Fanshawe College) - Kitchener - Toronto Pearson - Toronto.
Can't book any tickets, so it seems there are looking for a booking platform provider.

I can't comment on how it feels driving a Motor Coach for a line haul or charter as I'm not a driver, and can only comment as Bus Fan/passenger.
The longest trips I've taken by Motor Coach where:
Atlantic City to New York City 2 HRS(I think, it was over 20 years ago)
Transfer @ New York
New York City to Montreal 8 HRS I believe not sure 20 years ago
Montreal to Boston 8 HRS I think, it was some time ago. 2 times
A Charter 14 days Ottawa to New York State( a few places)-Vermont, Connetic(Hartford), Massatusses(Boston, Cape Cod, Mystic)

I took a look at the 6 coach companies serving the Ottawa-Toronto Toronto-Ottawa schedules to see what difference they offer. For Book-A-Ride the schedule was hard to find. I did not include stops between Ottawa & Toronto(IE Scarbourgh, Kingston etc) or price. From what I saw for price it vary from about $39 to $59.

Ex-McCoy H3-45 #224 is parked at the back of the Big Rig Collision Facility in S.E. Calgary with Alberta plates on it. You can see the former McCoy lettering.
